In their last scene together during Daryl Dixon season 2, episode 2, the couple talks about rescuing Laurent (Louis Puech Scigliuzzi) from the Nest and heading to America together. At the end of the conversation, they kiss, cementing the relationship that has developed between them since the beginning of the first season.
Talking to Entertainment Weekly, Reedus interpreted Daryl and Isabelle’s kiss in Daryl Dixon Season 2 as experimental instead of something specific. The actor explained that he felt his character questioned the idea of having to constantly fight, and instead wanted to try something less confrontational and more defined. I think it was an experiment. It was: “What is this?” It wasn’t “This is it!” It’s “What is this?” And what will it look like and what will it feel like? I don’t think there was any intention of getting to second base. I think it was more scary to pull back the curtains to look at the idea.
I’ll tell you my interpretation of this: he’s never had to do this before and he doesn’t know what it means. He’s been running and wrestling his whole life and asks himself: Did I do this wrong? Should I just find someone and live on a farm and just experience these moments before I die? Or should I fight and fight? He questions the whole big picture.
What Daryl and Isabelle’s kiss means for The Walking Dead: Daryl Dixon, Season 2
The couple’s relationship is likely to deepen in future episodes.

It means the duo will have to put their potential romance on hold while they work together to save him..
